The issue will be either permission related or the document root can not be found. You should receive a error message by directly going to the URL that Tile Pls is trying to load. For example:

http://localhost/wordpress/wp-content/plugins/connections/includes/libraries/timthumb/timthumb.php?src=http://localhost/wordpress/wp-content/connection_images/my_image.jpg

What is the error message?
